Application of a 5-bromo-2′-deoxyuridine ELISA for measuring the lymphoproliferative response to human cytomegalovirus in HIV-1-infected patients

2002

Assessment of the lymphoproliferative response to human cytomegalovirus (HCMV) may help to identify human immunodeficiency virus (HIV)-1-infected patients at high risk of developing HCMV end-organ disease. The tritiated thymidine ([3H]-TdR)-incorporation assay is the gold standard for measuring lymphoproliferative responses, though it is unsuitable as a routine laboratory procedure. An alternative non-radioactive technique, a 5-bromo-2'-deoxyuridine (BrdU) enzyme-linked immunosorbent assay, was applied for measuring T-cell proliferation in response to HCMV. Stimulation of either 1 x 10(5) or 5 x 10(4) peripheral blood mononuclear cells (PBMCs)/well with 10 PFU/well (before inactivation) of …

Presence of Epstein-Barr virus, cytomegalovirus and human papillomavirus in normal oral mucosa of HIV-infected and renal transplant patients

2001

OBJECTIVES: To determine the prevalence of EBV-DNA,CMV-DNA and HPV-DNA in oral healthy mucosa of HIV-infected and renal transplant patients. To associate the detection of viral genomes with laboratory parameters of immunodeficiency, gender, antiretroviral and immunosuppressive therapy. DESIGN: A cross-sectional analysis of lingual and buccal cytobrushings from HIV-infected and renal transplant patients. SUBJECTS AND METHODS: Lingual and buccal cytobrushings were obtained from clinically normal oral mucosa of 57 HIV+, 40 renal transplant patients and 30 healthy uninfected controls, all matched for age at baseline of examination. Presence of EBV-, CMV- and HPV-DNA was assessed by polymerase c…

Human conglutinin-like protein inhibits infection by the human immunodeficiency virus-1 in vitro.

1992

In summary the lectin-like protein analogous to bovine conglutinin was purified from human serum. Using a lectin-based ELISA system, it was demonstrated that conglutinin-like protein binds to human immunodeficiency virus-1 (HIV1) glycoprotein 120 (gp 120) via its carbohydrate binding site. In vitro experiments with T-lymphoblastoid CEM cells revealed that conglutinin-like protein abolishes infection by HIV1; a 50 % cytoprotective concentration of 23.9 μg/ml was measured.

Changes in CD4+ cells’ miRNA expression following exposure to HIV-1

2010

Background: Micro RNAs (miRNAs) inhibit HIV-1 expression by either modulating host innate immunity or by directly interfering with viral mRNAs. Here, we investigated the miRNA profile that discriminates different classes of HIV-1-infected patients from multiple-exposed uninfected individuals. Methods: The expression levels of 377 miRNAs were selectively analysed in CD4+ cells isolated from whole blood of HIV-1 elite LTNP (eLTNP), naive, and multiply exposed uninfected individuals (MEUs). MiRNA extraction was performed by the mir Vana™ miRNA Isolation Kit (Ambion), and their expression was subsequently examined by real-time PCR-based arrays. The expression of miRNAs was also determined in pr…

Flow cytometric investigation of neutrophil oxidative burst and apoptosis in physiological and pathological situations

2009

Flow cytometric analysis provides a rapid screen for abnormalities of polymorphonuclear neutrophils (PMN) function and reflect their behavior in vivo more accurately. This review summarizes the major fluorescent probes used to study PMN oxidative burst and apoptosis using flow cytometry (FCM). We also provide examples of FCM studies in physiological and pathological situations, illustrating the advantages of FCM for assessment of PMN oxidative burst and PMN apoptosis. These data point to the role of FCM in detecting primary immunodeficiencies such as IRAK4 deficiency and support the use of the assessment of the PMN oxidative burst for routine testing in patients with bacterial infections. W…

Human Immunodeficiency Virus Continuum of Care in 11 European Union Countries at the End of 2016 Overall and by Key Population: Have We Made Progress?

2020

Abstract Background High uptake of antiretroviral treatment (ART) is essential to reduce human immunodeficiency virus (HIV) transmission and related mortality; however, gaps in care exist. We aimed to construct the continuum of HIV care (CoC) in 2016 in 11 European Union (EU) countries, overall and by key population and sex. To estimate progress toward the Joint United Nations Programme on HIV/AIDS (UNAIDS) 90-90-90 target, we compared 2016 to 2013 estimates for the same countries, representing 73% of the population in the region. Methods A CoC with the following 4 stages was constructed: number of people living with HIV (PLHIV); proportion of PLHIV diagnosed; proportion of those diagnosed …

Community-acquired respiratory virus lower respiratory tract disease in allogeneic stem cell transplantation recipient: Risk factors and mortality fr…

2018

Abstract Risk factors (RFs) and mortality data of community‐acquired respiratory virus (CARVs) lower respiratory tract disease (LRTD) with concurrent pulmonary co‐infections in the setting of allogeneic hematopoietic stem cell transplantation (allo‐HSCT) is scarce. From January 2011 to December 2017, we retrospectively compared the outcome of allo‐HSCT recipients diagnosed of CARVs LRTD mono‐infection (n = 52, group 1), to those with viral, bacterial, or fungal pulmonary CARVs LRTD co‐infections (n = 15, group 2; n = 20, group 3, and n = 11, group 4, respectively), and with those having bacterial pneumonia mono‐infection (n = 19, group 5). Overall survival (OS) at day 60 after bronchoalveol…

Adolescents' knowledge and attitudes concerning HIV infection and HIV-infected persons: how a survey and focus group discussions are suited for resea…

1999

The purpose of this article is to examine how two different corpora of material are suited for researching the sexuality of youth on the basis of material gathered via a structured questionnaire (N = 1183 response rate 87%) and via eight focus group discussions (FGDs) and to investigate the knowledge and opinions of adolescents at the age of 15 years about HIV infection and HIV-infected persons. Both boys and girls showed a good level of knowledge about HIV infection and AIDS. While their level of knowledge was good their attitude was that the threat of an HIV infection was not a personal issue. Furthermore negative attitudes to those having HIV/AIDS became more pronounced the more socially…

Mapping age- and sex-specific HIV prevalence in adults in sub-Saharan Africa, 2000–2018

2022

Abstract Background Human immunodeficiency virus and acquired immune deficiency syndrome (HIV/AIDS) is still among the leading causes of disease burden and mortality in sub-Saharan Africa (SSA), and the world is not on track to meet targets set for ending the epidemic by the Joint United Nations Programme on HIV/AIDS (UNAIDS) and the United Nations Sustainable Development Goals (SDGs). Precise HIV burden information is critical for effective geographic and epidemiological targeting of prevention and treatment interventions. Age- and sex-specific HIV prevalence estimates are widely available at the national level, and region-wide local estimates were recently published for adults overall. We…

Changes in hearing in 80-year-old people: a 10-year follow-up study

2004

The aims of this prospective 10-year longitudinal population study were to report changes in hearing in people aged 80 years at baseline, and to assess such changes by comparing results from both cross-sectional and longitudinal analyses. The study population comprised all residents of the city of Jyväskylä born in 1910 (n = 291). The subjects' hearing was tested on three occasions at 5-year intervals with the use of pure-tone audiometry, speech audiometry, and self-report on hearing difficulties. The results showed a significant deterioration in hearing sensitivity in both the longitudinal and cross-sectional assessments over the 10-year follow-up. However, the change in self-assessed hear…
